tTg New Generation - a novel approach to Celiac disease testing

Tissue Transglutaminase (tTg) has been identified as the major target antigen in endomysium in 1997 by Dieterich and Schuppan et.al. Over the last 10 years, gliadin testing has been replaced by tTg testing thus, celiac disease diagnosis has risen to a sensitivity and specificity level unknown before.

Even after this main serological discovery, a vast majority of celiac patients still remain undetected (ratio of known to unknown CD is 1 to 7 and there’s a mean delay on a CD diagnosis of 5 to 11 years.

Several theories about the “iceberg model” were published, and studies have shown that CD also exists in different underestimated forms referred to as potential, latent and silent CD.

Meta-analyses of current studies indicate that celiac disease affects 1% of the population worldwide. Risk groups (like DM type 1 and Osteoporosis) have an even higher prevalence (from 1:20 to 1:7).

AESKULISA® tTg New Generation now sets new standards in sensitivity and specificity of celiac disease diagnosis.

The results of a recent Multi-Center Celiac Serology Study undertaken to define the optimal serological screening approach to Celiac Disease clearly showed that dual or combined isotype (IgA+IgG) transglutaminase screening can be advocated as a sensitive and specific alternative to endomysial antibody testing in the assessment of Celiac Disease. AESKULISA® tTg ELISA kits performed excellent. AESKULISA® Celicheck even placed first as the best dual isotype transglutaminase kit of all assays tested.

The reason is obvious: AESKULIA® tTg New Generation ELISA kits show a unique composition. Human recombinant tissue transglutaminase is cross-linked with gliadin-specific peptides resulting in the creation of neo-epitopes of tTg. The tests are highly specific as no cross-reactions with gliadin occur.

Thus the AESKULIA® tTg New Generation tests are the only available tTg test to mimic the physiologically relevant tTg enzyme complex – offering a sensitivity and specificity level unknown before.

Features of our tTg New generation kits are:

  • For detection of tissue transglutaminase antibodies, a new generation of tTg tests is available: by presenting neo-epitopes of tTg, the sensitivity of the tests could be markedly increased. Neo epitopes of tTg are induced by adding gliadin-specific peptides.

  • At the same time, the tests are highly specific as no cross-reactions with gliadin occur.

  • In addition, the new generation tTg assays employ human recombinant tissue transglutaminase, which ensures maximum specificity and sensitivity as compared to the guinea pig antigen, which shares only 80% homology with its human counterpart.
